Chromosome polymorphism in the Brazilian dwarf brocket deer, Mazama nana (Mammalia, Cervidae) Genet. Mol. Biol.
Abril,Vanessa Veltrini; Duarte,José Maurício Barbanti.
The Brazilian dwarf brocket deer (Mazama nana) is the smallest deer species in Brazil and is considered threatened due to the reduction and alteration of its habitat, the Atlantic Rainforest. Moreover, previous work suggested the presence of intraspecific chromosome polymorphisms which may contribute to further population instability because of the reduced fertility arising from the deleterious effects of chromosome rearrangements during meiosis. We used G- and C-banding, and nucleolus organizer regions localization by silver-nitrate staining (Ag-NOR) to investigate the causes of this variation. Mazama nana exhibited eight different karyotypes (2n = 36 through 39 and FN = 58) resulting from centric fusions and from inter and intraindividual variation in...

Comparative karyotype analysis in diploid and triploid Dolichoplana carvalhoi (Tricladida, Terricola, Rhynchodemidae) from Brazil Genet. Mol. Biol.
Alvarez,Luciana; Almeida,Eunice Judith Cardoso de.
In this work, we present cytogenetic data for the land planarian Dolichoplana carvalhoi. Two different karyotypes, one diploid (2n = 2x = 14 chromosomes) and one triploid (2n = 3x = 21 chromosomes), corresponding to two morphological body patterns, are described. Chromosomes from regenerating blastema were studied after routine Giemsa staining and CBG banding. Our analyses revealed heteromorphisms in chromosomes 2, 3 and 4 of the diploid karyotype and in chromosomes 1, 3 and 7 of the triploid karyotype. Further studies are needed in order to determine if the two morphological patterns of D. carvalhoi represent distinct species.

Cytogenetic damage related to low levels of methyl mercury contamination in the Brazilian Amazon Anais da ABC (AABC)
AMORIM,MARÚCIA I. M.; MERGLER,DONNA; BAHIA,MARCELO O.; DUBEAU,HÉLÈNE; MIRANDA,DANIELA; LEBEL,JEAN; BURBANO,ROMMEL R.; LUCOTTE,MARC.
The mercury rejected in the water system, from mining operations and lixiviation of soils after deforestation, is considered to be the main contributors to the contamination of the ecosystem in the Amazon Basin. The objectives of the present study were to examine cytogenetic functions in peripheral lymphocytes within a population living on the banks of the Tapajós River with respect to methylmercury (MeHg) contamination, using hair mercury as a biological indicator of exposure. Our investigation shows a clear relation between methylmercury contamination and cytogenetic damage in lymphocytes at levels well below 50 micrograms/gram, the level at which initial clinical signs and symptoms of mercury poisoning occur. The first apparent biological effect with...

Chromosome mapping of ribosomal genes and histone H4 in the genus Radacridium (Romaleidae) Genet. Mol. Biol.
Anjos,Allison; Loreto,Vilma; Souza,Maria José de.
In this study, two species of Romaleidae grasshoppers, Radacridium mariajoseae and R.nordestinum, were analyzed after CMA3/DA/DAPI sequential staining and fluorescence in situ hybridization (FISH) to determine the location of the 18S and 5S rDNA and histone H4 genes. Both species presented karyotypes composed of 2n = 23, X0 with exclusively acrocentric chromosomes. CMA3+ blocks were detected after CMA3/DA/DAPI staining in only one medium size autosome bivalent and in the X chromosome in R. mariajoseae. On the other hand, all chromosomes, except the L1 bivalent, of R. nordestinum presented CMA3+ blocks. FISH analysis showed that the 18S genes are restricted to the X chromosome in R. mariajoseae, whereas these genes were located in the L2,S9 and S10...

Comparative cytogenetic studies of Bufo ictericus, B. paracnemis (Amphibia, Anura) and an intermediate form in sympatry Genet. Mol. Biol.
Azevedo,MFC; Foresti,F.; Ramos,PRR; Jim,J.
Specimens of Bufo ictericus, Bufo paracnemis and a third type, considered an intermediate subgroup between these species, were cytogenetically studied by conventional Giemsa staining, C-banding and staining of the nucleolus organizer region (NOR). The nuclear DNA content and seroproteins were also analyzed to characterize these species, and verify the possibility of hybridization between them. Karyotypes and cytogenetic markers were essentially equal on the basis of the methods used. The DNA nuclear content found was 6.25 ± 0.30 pg/DNA in Bufo ictericus; 7.57 ± 0.40 pg/DNA in Bufo paracnemis and 7.04 ± 0.29 pg/DNA in the intermediate subgroup. Eletrophoresis of total blood serum in Bufo ictericus, Bufo paracnemis and the intermediate specimens revealed a...

Nucleolar organizer regions in Sittasomus griseicapillus and Lepidocolaptes angustirostris (Aves, Dendrocolaptidae): evidence of a chromosome inversion Genet. Mol. Biol.
Barbosa,Marcelo de Oliveira; Silva,Rubens Rodrigues da; Correia,Vanessa Carolina de Sena; Santos,Luana Pereira dos; Garnero,Analía del Valle; Gunski,Ricardo José.
Cytogenetic studies in birds are still scarce compared to other vertebrates. Woodcreepers (Dendrocolaptidae) are part of a highly specialized group within the Suboscines of the New World. They are forest birds exclusive to the Neotropical region and similar to woodpeckers, at a comparable evolutionary stage. This paper describes for the first time the karyotypes of the Olivaceous and the Narrow-billed Woodcreeper using conventional staining with Giemsa and silver nitrate staining of the nucleolar organizer regions (Ag-NORs). Metaphases were obtained by fibular bone marrow culture. The chromosome number of the Olivaceous Woodcreeper was 2n = 82 and of the Narrow-billed Woodcreeper, 2n = 82. Ag-NORs in the largest macrochromosome pair and evidence of a...

Chromosomal basis for a bilateral gynandromorph in Pycnogaster inermis (Rambur, 1838) (Orthoptera, Tettigoniidae) Naturalis
Barranco, P.; Cabrero, J.; Camacho, J.P.M.; Pascual, F..
Morphological and cytogenetic analyses of a spontaneous gynandromorph of Pycnogaster inermis, found in a natural population in the Spanish Sierra Nevada, have shown that the left side was male and the right side female, with almost perfect bilateral symmetry. Ventral valves of the ovipositor were very short, appearing as two spurs; dorsal valves were longer but still shorter than normal. The specimen had an apparently normal testis on the left side and a well-developed ovary on the right side. Cytogenetic analysis revealed the presence of 2n = 28 + X0 chromosomes in the testis and 2n = 28 + XX in the ovary, the same as standard males and females, respectively, of this species. This suggests that the gynandromorphism occurred at a very early developmental...

Study of genetic damage in the japanese oyster induced by an environmentally-relevant exposure to diuron: evidence of vertical transmission of dna damage ArchiMer
Barranger, Audrey; Akcha, Farida; Rouxel, Julien; Brizard, Raphael; Maurouard, Elise; Pallud, Marie; Menard, Dominique; Tapie, N.; Budzinski, Helene; Burgeot, Thierry; Benabdelmouna, Abdellah.
Pesticides represent a major proportion of the chemical pollutants detected in French coastal waters and hence a significant environmental risk with regards to marine organisms. Commercially-raised bivalves are particularly exposed to pollutants, among them pesticides, as shellfish farming zones are subject to considerable pressure from agricultural activities on the mainland. The aims of this study were to determine (1) the genotoxic effects of diuron exposure on oyster genitors and (2) the possible transmission of damaged DNA to offspring and its repercussions on oyster fitness. To investigate these points, oysters were exposed to concentrations of diuron close to those detected in the Marennes-Oleron Basin (two 7-day exposure pulses at 0.4 and 0.6 μg...

Barth,Adriane; Fernandes,Anderson; Pompolo,Silvia das Graças; Costa,Marco Antônio.
Tetragonisca angustula and Tetragonisca fiebrigi have recently been listed as valid species. This study aimed to cytogenetically investigate both species, emphasizing the new registry of B chromosomes in the tribe Meliponini. We analyzed colonies of T. angustula and T. fiebrigi collected at Tangará da Serra, Mato Grosso, Brazil, through conventional Giemsa staining, C-banding, and base-specific fluorochrome staining (CMA3/DAPI). T. angustula showed 2n = 34 chromosomes in females and n = 17 in males, with karyotype formula 2K = 34A M. T. fiebrigi showed numeric variation, with chromosome number varying from 2n = 34 to 2n = 36 in females and from n = 17 to n=18in males, with karyotype formula 2K = 32A M+2A Mc and 2K = 32A M+2A Mc + 1 or 2 B-chromosomes. Cytotaxonomy of Parodon nasus and Parodon tortuosus (Pisces, Characiformes): A case of synonymy confirmed by cytogenetic analyses Genet. Mol. Biol.
Bellafronte,Elisangela; Margarido,Vladimir Pavan; Moreira-Filho,Orlando.
Morphological and cytogenetical studies were carried out on the freshwater fish Parodon nasus and Parodon tortuosus in order to evaluate a putative synonymy. The diploid chromosome number observed in both species was 2n = 54 (48M/SM and 6ST) with no differences between the sexes. Despite slight differences in the pattern of heterochromatin distribution and the number of cusps in symphysean teeth, the location of nucleolar organizer regions (NORs) and 5S rRNA genes (both species-specific features) were similar in both species. The remarkable similarity observed between these allopatric species supports recent taxonomical reviews indicating that P. tortuosus is a synonym for P. nasus.

DNA barcode of Parodontidae species from the La Plata river basin - applying new data to clarify taxonomic problems Neotropical Ichthyology
Bellafronte,Elisangela; Mariguela,Tatiane Casagrande; Pereira,Luiz Henrique Garcia; Oliveira,Claudio; Moreira-Filho,Orlando.
In the past years, DNA barcoding has emerged as a quick, accurate and efficient tool to identify species. Considering the difficulty in identifying some Parodontidae species from the La Plata basin and the absence of molecular data for the group, we aimed to test the effectiveness of DNA barcoding and discuss the importance of using different approaches to solve taxonomic problems. Eight species were analyzed with partial sequences of Cytochrome c oxidase I. The mean intraspecific K2P genetic distance was 0.04% compared to 4.2% for mean interspecific K2P genetic distance. The analyses of distance showed two pairs of species with K2P genetic divergence lower than 2%, but enough to separate these species. Apareiodon sp. and A. ibitiensis, considered as the...

Sex chromosome system ZZ/ZW in Apareiodon hasemani Eigenmann, 1916 (Characiformes, Parodontidae) and a derived chromosomal region Genet. Mol. Biol.
Bellafronte,Elisangela; Schemberger,Michelle Orane; Artoni,Roberto Ferreira; Moreira Filho,Orlando; Vicari,Marcelo Ricardo.
Parodontidae fish show few morphological characteristics for the identification of their representatives and chromosomal analyses have provided reliable features for determining the interrelationships in this family. In this study, the chromosomes of Apareiodon hasemani from the São Francisco River basin, Brazil, were analyzed and showed a karyotype with 2n = 54 meta/submetacentric chromosomes, and a ZZ/ZW sex chromosome system. The study revealed active NORs located on pair 11 and additional 18S rDNA sites on pairs 7 and 22. The 5S rDNA locus was found in pair 14. It showed a pericentric inversion regarding the ancestral condition. The satellite DNA pPh2004 was absent in the chromosomes of A. hasemani, a shared condition with most members of Apareiodon....

Identification of distinct evolutionary units in allopatric populations of Hypostomus cf. wuchereri Günther, 1864 (Siluriformes: Loricariidae): karyotypic evidence Neotropical Ichthyology
Bitencourt,Jamille de Araújo; Affonso,Paulo Roberto Antunes de Mello; Giuliano-Caetano,Lucia; Dias,Ana Lucia.
Few chromosomal reports are available for the endemic fish fauna from coastal basins in northeastern Brazil, and regional biodiversity remains partially or completely unknown. This is particularly true for Loricariidae, the most diverse family of armored catfishes. In the present work, allopatric populations of Hypostomus cf. wuchereri (Siluriformes: Loricariidae) from two basins in Bahia (northeastern Brazil) were cytogenetically analyzed. Both populations shared 2n = 76 chromosomes, a karyotype formula of 10m+18sm+48st/a (FN = 104) and single terminal GC-rich NORs on the second metacentric pair. Nevertheless, microstructural differences were detected by C-banding, fluorochrome staining and chromosomal digestion with restriction enzymes (Alu I, Bam HI,...

Karyotypic diversity between allopatric populations of the group Hoplias malabaricus (Characiformes: Erythrinidae): evolutionary and biogeographic considerations Neotropical Ichthyology
Blanco,Daniel Rodrigues; Lui,Roberto Laridondo; Bertollo,Luiz Antonio Carlos; Margarido,Vladimir Pavan; Moreira Filho,Orlando.
Three populations of the group Hoplias malabaricus from the hydrographic basins of the São Francisco, Araguaia/Tocantins and Xingu Rivers in Brazil were analyzed using classic cytogenetic methods (Giemsa staining, C-banding and Ag-NORs) and molecular methods (fluorescent in situ hybridization with 18S rDNA, 5S rDNA and 5SHindIII satellite DNA probes). The chromosome markers allowed the characterization of these populations as belonging to karyomorph A and the detection of inter-population divergences. These differences likely stem from different evolutionary histories resulting from geographic isolation between populations associated to the dispersive mode of these organisms, reinforcing genetic diversity in the group Hoplias malabaricus.

New contributions to the study of Corixoidea: cytogenetic characterization of three species of Sigara from Argentina and the plausible mechanisms of karyotype evolution within Nepomorpha Rev. Soc. Entomol. Argent.
Bressa,María José; Papeschi,Alba Graciela.
Cytogenetic studies in Heteroptera contribute to the analysis of evolutionary trends within the group. Heteroptera are characterized by the possession of holokinetic chromosomes, different sex chromosome mechanisms and a pair of m chromosomes in some species. In the present work, the male karyotype and meiosis in Sigara denseconscripta (Breddin), S. chrostowskii Jaczewski, and S. rubyae (Hungerford) are described. The three species share a diploid chromosome number of 2n= 24 with a pair of m chromosomes and an XY/XX sex chromosome system. With this study the chromosome number of 30 species of Corixoidea are known and the modal karyotype is 2n= 20+2m+XY in males. The available cytogenetic information in Heteroptera led us to suggest that the presence of a...

Chromosome comparison between populations of the collared peccary, Tayassu tajacu, raised in captivity Biocell
Carvalho de Souza,Patrícia; Salim Khayat,André; Chamon Seligmann,Igor; Rodríguez Burbano,Rommel Mario.
The collared peccary (Tayassu tajacu) is widely distributed over the American continent, being found from the south of the USA to the north of Argentina. In Brazil, it is spread all over the country, being one of the potential species to be raised in captivity. Therefore, the cytogenetic techniques could be a potential tool for reproductive monitoring of animals raised in captivity, mainly when destined for commercial purposes. This study had the objective of determining the chromosome number of two populations raised in captivity and characterizing them by GTG banding. For this purpose, an analysis was made of mitotic metaphases obtained from lymphocyte cultures made from blood samples of 11 animals, six of which from the Northeast and five from the North...

Chromosomal analyses in Megalonema platanum (Siluriformes: Pimelodidae), an endangered species from South American rivers Neotropical Ichthyology
Carvalho,Rafael Augusto de; Sanchez,Sebástian; Swarça,Ana Claudia; Fenocchio,Alberto Sergio; Martins-Santos,Isabel C.; Dias,Ana Lúcia.
This study presents chromosomal data of Megalonema platanum from rio Tibagi, Paraná, Brazil and from rio Paraná, Argentina. The diploid number was equal 54 with karyotype composition of 24m+16sm+2st+12a in both populations. The AgNOR sites were detected in the terminal position of a submetacentric pair of the two analyzed populations, coinciding with secondary constrictions on the short arm of pair 15. CMA3 and FISH with 18S rDNA probe displayed fluorescent signals that correspond to the AgNOR sites and secondary constriction. The presence of a small acrocentric supernumerary chromosome can be observed in M. platanum from rio Tibagi, with centromeric heterochromatin. Others heterochromatic blocks were evidenced in the terminal position of some chromosome...

Meiotic chromosomes and nucleolar behavior in testicular cells of the grassland spittlebugs Deois flavopicta, Mahanarva fimbriolata and Notozulia entreriana (Hemiptera, Auchenorrhyncha) Genet. Mol. Biol.
Castanhole,Márcia Maria Urbanin; Pereira,Luis Lênin Vicente; Souza,Hederson Vinicius de; Valério,José Raul; Barbosa,Leonardo Rodrigues; Itoyama,Mary Massumi.
Spittlebugs annually infest pastures and cause severe damage, representing a serious problem for the tropical American beef cattle industry. Spittlebugs are an important biotic constraint to forage production and there is a lack of cytogenetic data for this group of insects. For these reasons, we conducted this work, in which the spermatogenesis and nucleolar behavior of Deois flavopicta, Mahanarva fimbriolata and Notozulia entreriana were studied. The males possessed testes in the shape of a "bunch of grapes"; a variable number of testicular lobes per individual and polyploid nuclei composed of several heteropycnotic bodies. A heteropycnotic area was located in the periphery of the nucleus (prophase I); the chiasmata were terminal or interstitial;...
